How to use the cleaning verification swab?

1. Select the appropriate cleaning verification swab.

At present, chemical sampling and microbial sampling for cleaning verification in pharmaceutical workshops require wiping cotton swabs. There are two main types in terms of shape. They are all one-piece cotton swabs. One of them is a smooth sampling rod, and the other is close to the sampling head. There is an easy fold opening.

The first kind of sampling cotton swab needs to use scissors to cut the sampling rod after sampling is completed before putting it into the glass test tube. So as to prevent the part touched by the hand from interfering with the microbiological or chemical results.

In the process of using scissors to cut the sampling rod, there is a risk of microbial contamination caused by incomplete cleaning of the scissors, resulting in false positive results, resulting in unqualified cleaning verification results. Therefore, choosing a cleaning verification swab with a breakpoint can reduce the risk of sample contamination.

2. Do a wipe

(1) Wipe the area so that the amount of residue conforms to the linear range of the detection method.

Usually, 25cm² or 100cm² can be used to determine the wipe solvent and swab, and specify the degree of wetness of the swab. For example, a swab is moistened and squeezed over a solvent bottle until no dripping occurs to collect samples from certain areas inside a pharmaceutical production tank. These areas can be the inner wall of the tank or a small plate called a “specimen” (during production During the process, this small plate floats on the surface of the product and acts as a marker of how clean and cleaned the production tank is). A cotton swab is placed on the surface to be sampled, and the entire surface of the sample is wiped. The direction and pressure of wiping are defined in the Cleaning Standard Operating Procedure (SOP).

(2) Wiping skills

When wiping, press the tip of the cotton swab on the sampling surface, force it to bend it at 45° to the wiping surface, wipe the sampling surface smoothly and slowly, and move it from one side to the other while moving forward. Cover the entire surface. Turn the cotton swab over and wipe the other side of the cotton swab, but it is perpendicular to the moving direction of the previous wiping. Each cotton swab is wiped and sampled 25cm² separately. The sampling position should not be repeated with the microbial sampling point.

After wiping, cut the cotton swabs from each sampling point and put them in the same sample receiving tube, and seal them with stoppers or caps.

3. Extract the sampled cotton swab

Put a cleaning swab into a vial containing an acidic solution, then perform sonication or centrifugation, and finally analyze the TOC concentration of the acidic solution. The swab extract can also be analyzed by placing the swab in a sealed glass ampoule containing 5% phosphoric acid and 100 g/L sodium persulfate solution. Carbon dioxide in the form of inorganic carbon is purged to the environment, and the glass bottle is then sealed. Glass vials are heated to 100°C and stored for the specified time. Finally, when the glass bottle is opened, TOC is blown out in the form of carbon dioxide and detected.

Notice:

• The tools and containers used for sampling shall be sterilized;
• After the sampling is completed, a label should be attached to the sample tube, including sampling location, sampling purpose, sampling number, sampling person and sampling date, etc.
